Si chiama C. elegans, ed è un piccolo invertebrato che ha permesso ai ricercatori del Cnr di analizzare, in vivo, gli eventi che precedono la morte neuronale alla base di malattie come la Sma
L’atrofia muscolare spinale (Sma) è una malattia neurodegenerativa rara, ma tra le più comuni cause di mortalità infantile, colpendo un neonato ogni 6.000-10.000 in tutto il mondo. È caratterizzata dalla morte di alcuni neuroni motoridel midollo spinale ed è causata dalla mancanza del gene Smn1. Non esistendo ancora una terapia efficace, sono di fondamentale importanza gli studi di ricerca di base su questo gene e l’utilizzo di piccoli organismi modello. Ricercatori dell’Istituto di bioscienze e biorisorse del Consiglio nazionale delle ricerche (Ibbr-Cnr) di Napoli hanno analizzato, per la prima volta, gli eventi iniziali che accadono durante la morte neuronale quando manca il gene Smn1. La ricerca è pubblicata sulla rivista Human Molecular Genetics.
“I nostri studi ci hanno permesso di dimostrare che la mancanza di Smn1 scatena un meccanismo di morte programmata, chiamata apoptosi”, spiega Elia Di Schiavi dell’Ibbr-Cnr, morte che, però, può essere contrastata in vari modi. “Abbiamo dimostrato l’intercambiabilità tra il gene umano e quello diCaenorhabditis elegans, un piccolo animale invertebrato che costituisce un importante modello genetico per l’analisi dei processi neuro-degenerativi: attraverso la sostituzione del gene di C. elegans con quello umano siamo riusciti infatti a prevenire la morte dei neuroni. Inoltre, è stato possibile prevenire la neuro-degenerazione utilizzando un farmaco, l’acido valproico, già impiegato in trials clinici su pazienti affetti da Sma. Studiare quello che succede in C. elegans ha quindi un impatto ed è rilevante anche per capire la funzione del gene umano, la cui mancanza provoca la Sma”.
Lo studio mostra però altri modi per evitare la neuro-degenerazione nel C. elegans. “Per prevenire la morte neuronale gioca un ruolo importante anche un altro gene, Plastin3, la cui presenza permette in parte di scongiurare la morte dei neuroni”, prosegue il ricercatore dell’Ibbr-Cnr. “Questi esperimenti spiegano a livello molecolare il dato clinico secondo cui i pazienti che dispongono di maggiori quantità di Plastin3 hanno una forma meno grave della malattia”.
I ricercatori hanno utilizzato questo modello animale per le sue caratteristiche, che lo rendono ottimale per valutare le conseguenze di mutazioni genetiche sulla sopravvivenza dei neuroni. “C. elegans, seppur evolutivamente distantissimo dall’uomo, possiede molti geni che svolgono una funzione simile nell’essere umano”, conclude Di Schiavi. “La nostra ricerca ci ha permesso di scoprire che tra questi geni che hanno una conservazione funzionale c’è anche il gene Smn1”.

Spinal muscular atrophy (SMA) is a rare neurodegenerative disorder, but among the most common causes of infant mortality, affecting one baby every 6,000-10,000 worldwide. It is characterized by the death of some neurons motoridel spinal cord and is caused by the absence of the SMN1 gene. As there is still an effective therapy, it is of fundamental importance for basic research studies of this gene and the use of small model organisms. Researchers at the Institute of Biosciences and bioresources of the National Research Council (IBBR-CNR) in Naples analyzed, for the first time, the initial events that occur during neuronal death when missing the SMN1 gene. The research is published in the journal Human Molecular Genetics.
"Our studies have allowed us to demonstrate that the lack of SMN1 triggers a programmed death mechanism called apoptosis," explains Elia Di Schiavi dell'Ibbr-CNR, death, however, can be countered in various ways. "We have demonstrated the interchangeability between the human gene and the diCaenorhabditis elegans, a small invertebrate animal which constitutes an important genetic model for the analysis of neuro-degenerative processes: through the replacement of the C. elegans gene with the human we managed fact to prevent the death of neurons. Moreover, it was possible to prevent the neuro-degeneration using a drug, valproic acid, already employed in clinical trials of patients with Sma. Study what happens in C. elegans has therefore an impact and it is also important to understand the human gene function, the lack of which causes the Sma ".
The study shows, however, other ways to prevent neuro-degeneration in C. elegans. "To prevent neuronal cell death plays an important role in another gene, Plastin3, whose presence allows in part to avert the death of neurons", says the researcher dell'Ibbr-Cnr. "These experiments explain the clinical data at the molecular level that patients who have higher amounts of Plastin3 have a less severe form of the disease."
Researchers have used this animal model because of its characteristics, which make it optimal for assessing the consequences of genetic mutations on the survival of neurons. "C. elegans, although evolutionarily distantissimo man, has many genes that play a similar role in humans, "concludes Di Schiavi. "Our research has enabled us to discover that among these genes that have a functional conservation there is also the SMN1 gene."
