Ricercatori In-Cnr dimostrano per la prima volta il ruolo di una proteina nel sistema nervoso centrale. I loro risultati aprono la strada a possibili innovativi trattamenti per i pazienti affetti da queste patologie.
Epilessia e disabilità intellettiva, patologie che si manifestano spesso in età infantile, sono in alcuni casi causate da mutazioni genetiche riguardanti il gene Kiaa1202, che contiene le informazioni per produrre la proteina Shrm4. Uno studio internazionale pubblicato sulla rivista Nature Communication, coordinato da Maria Passafaro dell’Istituto di neuroscienze del Consiglio nazionale delle ricerche di Milano (In-Cnr) ha dimostrato uno dei meccanismi attraverso cui tali mutazioni potrebbero causare queste patologie.
“Il mondo della ricerca è da sempre impegnato nel tentare di capire la relazione che intercorre fra mutazioni genetiche e l’insorgere di queste malattie e da recenti studi è stata riscontrata una stretta relazione tra famiglie che presentano tali patologie e mutazioni del gene Kiaa1202”, spiega Passafaro. “Il nostro studio ha dato un contributo ulteriore alla comprensione di queste patologie, scoprendo il ruolo svolto dalla proteina Shrm4. Questa proteina è molto importante in quanto responsabile del corretto posizionamento del recettore GabaB. Abbiamo osservato infatti che tale fenomeno è mediato dal motore molecolare dineina, e Shrm4 agisce da adattatore fra il motore e il recettore GabaB, permettendone la corretta localizzazione nelle sinapsi (punti di contatto che consentono la comunicazione dei neuroni tra loro e con altre cellule). Ciò che è stato scoperto è che quando Shrm4 è assente il recettore GabaB non riesce a raggiungere le sinapsi e non svolge quindi la propria funzione”.
I ricercatori hanno infatti registrato le correnti nell’ippocampo di animali privati di Shrm4, scoprendo una riduzione nella loro componente inibitoria. “La conseguenza è un aumento di crisi epilettiche in questi animali, insieme ad altri difetti quali aumento di comportamenti ansiosi, difficoltà di apprendimento e nella socialità. Da questi risultati potremo partire per mettere a punto possibili trattamenti per i pazienti affetti da mutazioni in tale gene”, conclude Passafaro.

Epilepsy and intellectual disability, pathologies often manifested in childhood, are in some cases caused by genetic mutations concerning the Kiaa1202 gene, which contains information to produce the Shrm4 protein. An international study published in the journal Nature Communication, coordinated by Maria Passafaro of the Institute of Neuroscience of the National Research Council of Milan (In-Cnr), showed one of the mechanisms through which these mutations could cause these pathologies.
"The research world has always been engaged in trying to understand the relationship between genetic mutations and the emergence of these diseases and recent studies has been found to have a close relationship between families presenting such pathologies and mutations in the Kiaa120 gene, Explains Passafaro. "Our study has further contributed to the understanding of these pathologies by discovering the role played by Shrm4 protein. This protein is very important as it is responsible for the correct positioning of the GabaB receptor. We have observed that this phenomenon is mediated by the molecular dinein engine, and Shrm4 acts as an adapter between the engine and the GabaB receptor, allowing it to locate correctly in the synapses (contact points that allow neurons to communicate with each other and with other cells). What has been discovered is that when Shrm4 is absent the GabaB receptor can not reach the synapses and therefore does not function. "
Researchers have recorded the currents in the Shrm4 private animal hippocampus, finding a reduction in their inhibitory component. "The consequence is an increase in epileptic seizures in these animals, along with other defects such as increased anxiety, learning difficulties, and sociality. From these results we can start to work out treatments for patients with mutations in that gene, "concludes Passafaro.
