È l’obiettivo del progetto Chimera che ha portato i ricercatori del Dipartimento di Fisica del Politecnico di Milano al brevetto di un innovativo strumento in grado di misurare la chiralità di farmaci e proteine.


Presto la aziende farmaceutiche potrebbero avere a disposizione un nuovo dispositivo per la misura della chiralità di farmaci e proteine. Lo strumento sviluppato all’interno del progetto Chimera (Chiral Measurements by Heterodyne Amplification) è al momento ancora in fase di sviluppo tra i muri del laboratorio del Dipartimento di Fisica del Politecnico di Milano, dove i ricercatori hanno già ottenuto il brevetto per l’innovativo dispositivo.
Una molecola è chirale se in natura esiste sotto due forme che, pur con la stessa formulazione chimica, si dispongono secondo strutture geometriche specularmente opposte. In un farmaco le due forme chirali della stessa molecola possono dare effetti diversi sull’organismo. È il caso tragico, per esempio, della talidomide, farmaco molto utilizzato negli anni ’50, che solo in seguito si scoprì essere teratogeno in una delle due forme chirali. Se assunta dalle donne incinte, infatti, una delle due forme speculari possedeva i voluti effetti ansiolitici e sedativi, mentre l’altra provocava gravi malformazioni ai feti. La caratterizzazione della chiralità delle molecole è di fondamentale importanza anche nello studio della struttura di alcune proteine, dal cui comportamento dipendono malattie degenerative come l’Alzheimer e il Parkinson.
Obiettivo finale del progetto Chimera è arrivare all’ingegnerizzazione e commercializzazione di uno strumento per la misura della chiralità che presenta molti vantaggi in termini di compattezza, semplicità di funzionamento, velocità di misura e quantità di informazioni misurabili rispetto ai prodotti commerciali disponibili sul mercato. Il nuovo dispositivo permetterà, inoltre, di abbattere i costi di produzione e il prezzo di vendita, consentendo la crescita delle dimensioni del mercato.
Lo strumento ha già dato ottimi risultati e a lungo termine potrà entrare a far parte dei processi di controllo qualità delle aziende farmaceutiche, nonché diventare parte di uno strumento diagnostico clinico. Per questo si è già meritato il Premio Research Ideas for the Market, tenutosi a Catania durante la conferenza Materials 2016. Mentre Dario Polli, il principal Investigator ha ottenuto un prestigioso finanziamento dall’European Research Council denominato “ERC-Proof of Concept” di 150.000 euro in 1 anno per portare l’idea fuori dai laboratori e immetterla sul mercato.

It is the goal of the Chimera project that has led the researchers of the Department of Physics of the Politecnico of Milan to patent an innovative tool that can measure the chirality of drugs and proteins.

Soon, pharmaceutical companies may have a new device for measuring chirality of drugs and proteins. The instrument developed within the Chimera (Chiral Measurements by Heterodyne Amplification) project is currently under development from the walls of the Department of Physics at the Politecnico di Milano, where researchers have already obtained the patent for the innovative device.
A molecule is chiral if it exists in nature under two forms which, although with the same chemical formulation, are disposed of by specularly opposite geometric structures. In a drug, the two chiral forms of the same molecule can have different effects on the body. It is the tragic case, for example, of thalidomide, a drug widely used in the 1950s, which only later found itself teratogenic in one of two chiral forms. If taken by pregnant women, one of the two specular forms possessed the desired anxiolytic and sedative effects, while the other caused serious malformations to the fetuses. The characterization of the chirality of molecules is of crucial importance also in the study of the structure of some proteins, whose behavior depends on degenerative diseases such as Alzheimer's and Parkinson's.
The ultimate goal of the Chimera project is to arrive at the engineering and marketing of a chirality measurement instrument that has many advantages in terms of compactness, ease of operation, the speed of measurement and measurable amount of information compared to commercial products available on the market. The new device will also reduce production costs and sales prices, allowing the market size to grow.
The instrument has already achieved great results and in the long run it can become part of quality control processes of pharmaceutical companies and become part of a clinical diagnostic tool. For this reason, the Research Ideas for the Market Prize, which was held in Catania during the Materials Conference 2016, has already been deserved. While Dario Polli, the principal Investigator has received a prestigious funding from the European Research Council called "ERC-Proof of Concept" of 150,000 Euro in 1 year to bring the idea out of the labs and enter it on the market.
