E' possibile stabilire l'ora esatta del decesso di una persona analizzando campioni cellulari di quattro tessuti per rilevare i livelli di attività residua dell'RNA dopo la morte. La tecnica, che ha un margine di incertezza di soli nove minuti, potrà essere di grande aiuto ai medici legali impegnati in indagini forensi.

I cambiamenti nell'espressione dei geni che avvengono nei tessuti dopo la morte possono essere usati per stabilire con esattezza il momento del decesso se i campioni di tessuto sono prelevati entro una trentina di ore.

La scoperta – frutto di uno studio condotto da ricercatori delle Università di Barcellona, in Spagna, e di Porto, in Portogallo, e pubblicato su “Nature Communications” - mette a disposizione dei medici legali impegnati in indagini forensi un nuovo potente strumento.

A differenza del DNA, che è abbastanza stabile per lunghi periodi post-mortem, l'RNA è molto più labile e sensibile alla degradazione, ma finora si sapeva molto poco su come la morte e il periodo trascorso dal decesso influenzano questa degradazione e, quindi, il livello dell’espressione dei geni in quel lasso di tempo.

Pedro G. Ferreira  e colleghi hanno analizzato 7015 campioni di 36 differenti tessuti prelevati da 540 donatori deceduti di cui era nota l’ora del decesso, effettuando il sequenziamento dell’RNA (RNA-seq) a intervalli regolari.

L’analisi dei dati ha confermato che i tempi di degradazione dell’RNA – e quindi di blocco dell’attività di trascrizione dei geni – variano da tessuto a tessuto, andando da un minimo di 17 minuti a in massimo di 1739 minuti, circa 29 ore.

In tutti i tessuti, inoltre, il tasso di trascrizione ha rivelato uno schema specifico per ciascuno di essi, anche se nella maggioranza dei tessuti si osserva un picco di decrescita dell’attività di trascrizione intorno alle sei ore dal decesso.

I ricercatori hanno però scoperto che nelle ore immediatamente successive alla morte in diversi tessuti si osserva un iniziale aumento della trascrizione di vari geni, come quella delle globine 
alfa dell’emoglobina, coinvolte nel trasporto dell’ossigeno dai polmoni ai tessuti. In diversi tessuti si osserva inoltre un aumento della trascrizione di vari fattori di crescita dopo ben quattro ore dal decesso.

Prendendo in esame sistematicamente tutti questi dati i ricercatori sono stati in grado di definire un modello che permette di risalire dal profilo di attività/degrado dell’RNA al momento esatto della morte. Dato però che in uno scenario forense tipico è probabile che siano disponibili pochi tessuti, Ferreira e colleghi si sono quindi preoccupati di ottimizzare il modello, trovando che è possibile ottenere un margine di incertezza di appena nove minuti nella definizione dell'ora analizzando solo le cellule di quattro tessuti: la pelle, la tiroide, i polmoni e il tessuto adiposo sottocutaneo.

It is possible to establish the exact time of death of a person by analyzing cell samples of four tissues to detect levels of residual activity of RNA after death. The technique, which has a margin of uncertainty of only nine minutes, can be of great help to the legal doctors engaged in forensic investigations.

Changes in the expression of genes that occur in tissues after death can be used to accurately determine the time of death if tissue samples are taken within thirty hours.

The discovery - the result of a study conducted by researchers from the Universities of Barcelona, ​​Spain, and Porto, Portugal, and published in "Nature Communications" - provides a powerful new tool to the legal doctors involved in forensic investigations.

Unlike DNA, which is fairly stable for long post-mortem periods, RNA is much more labile and susceptible to degradation, but until now very little was known about how death and the period since death affect this degradation and, therefore, , the level of gene expression in that time frame.

Pedro G. Ferreira and colleagues analyzed 7015 samples of 36 different tissues taken from 540 deceased donors whose death hour was known, performing RNA sequencing (RNA-seq) at regular intervals.

The analysis of the data confirmed that the RNA degradation times - and therefore the blocking of gene transcription activity - vary from tissue to tissue, going from a minimum of 17 minutes to a maximum of 1739 minutes, about 29 hours.

Moreover, in all tissues, the transcription rate revealed a specific pattern for each of them, even if in the majority of tissues a decrease of the transcription activity is observed around six hours after death.

However, the researchers found that in the hours immediately following death in different tissues there is an initial increase in the transcription of various genes, such as that of the hemoglobin alpha globin, involved in the transport of oxygen from the lungs to the tissues. In several tissues there is also an increase in the transcription of various growth factors after a full four hours after death.

By examining all these data systematically, the researchers were able to define a model that allows to trace the RNA activity / degradation profile at the exact moment of death. However, given that in a typical forensic scenario it is likely that few tissues are available, Ferreira and colleagues have therefore been concerned with optimizing the model, finding that it is possible to obtain a margin of uncertainty of just nine minutes in defining the time by analyzing only the cells of four tissues: the skin, the thyroid, the lungs and the subcutaneous adipose tissue.
